MOTS-c

The exercise-in-a-bottle peptide. It builds brand-new mitochondria (your cells' power plants) and flips the same energy switch as a hard workout, turning your metabolism back to what it was 10 to 20 years ago.

Your starting dose
1.25 to 2.5units
0.25 to 0.5 mg

Every day, under the skin

1.25 to 2.5 units 050100 1 mL

Your insulin syringe is marked 0 to 100. That whole line is 1 mL. Draw to the 1.25 to 2.5 mark.

The protocolHow to mix it, how much to draw, and how long a vial lasts.
The numbers
Vial40 mg
Mix with2 mL BAC water → makes 1 mg = 5 units
Start1.25 to 2.5 units (0.25 to 0.5 mg), daily, for the first 1 to 2 weeks
Work up to5 units (1 mg), daily, the standard dose
Fat-loss range7.5 to 10 units (1.5 to 2 mg), daily
FrequencyEvery day, under the skin
WhenAM, with carbs on board, never fully fasted / low-carb
Cycle12 weeks
Units made simple: your insulin syringe is marked 0 to 100. That whole line = 1 mL. Start by drawing to the 2.5 mark (0.5 mg) the first weeks, then move to the 5 mark (1 mg). No math.
How long one bottle lasts
At 0.5 mg 2.5u~80 days
At 1 mg 5u~40 days
At 2 mg 10u~20 days
One 40 mg vial = 200 units total. At the standard 1 mg/day that's about 40 days a bottle, so a full 12-week cycle runs roughly 2 bottles.

What it doesWhat you get, how it does it, and what to know before you run it.
What it does for you
Builds NEW mitochondriaMost "energy" products just squeeze more out of the power plants you have. MOTS-c signals your body to build brand-new ones, Trevor cites ~300% rise in biogenesis markers over 4 weeks. More plants = more baseline energy.
More and better ATPNot just more energy, cleaner energy. It improves how efficiently your mitochondria turn oxygen into ATP, so you get more output with less waste (less of the "exhaust" that shows up as fatigue and inflammation).
"Exercise in a bottle"It flips the exact same AMPK switch a hard workout does, your body literally makes more MOTS-c when you train. It puts your cells into "I've been exercising, mobilize energy and burn fuel" mode.
Kills the late-day crashThat 3 to 4pm wall reframed: it's your mitochondria running low, not your age. MOTS-c rebuilds the engine that's supposed to carry you through the day.
Better metabolism and recompIt shuttles carbs into muscle instead of fat (nutrient partitioning) and improves insulin sensitivity, the mechanism behind "lose fat, hold muscle."
How it works

MOTS-c is a 16-amino-acid peptide your body already makes, coded inside your mitochondrial DNA. It's a "retrograde signal": made in the mitochondria, it travels back to the cell's nucleus and changes which genes turn on. It flips the master energy switch AMPK → PGC-1α, which does two things: it drives mitochondrial biogenesis (builds new power plants) and clears out the broken ones. It also drives glucose into muscle without needing insulin, a "glucose vacuum" that fuels the muscle instead of feeding fat. And because MOTS-c expression naturally falls as you age, restoring it isn't forcing a drug on your body, it's topping up a signal you've been getting less of every year. The net result is the same message a hard workout sends: more energy, better fat-burning, sharper metabolism.

What to know before you run it
It's inherited only from your mom. MOTS-c is coded in your mitochondrial DNA, and you get 100% of your mitochondria from your mother. So this is a peptide passed down your maternal line, literally written into the DNA she gave you.
Your body makes MORE of it when you exercise. MOTS-c levels rise with a hard workout, it's one of the molecules that carries out the benefits of exercise. That's why it's nicknamed "exercise in a bottle."
Discovered at USC in 2015. It was identified by researchers at USC and published in Cell Metabolism, a hidden gene inside the mitochondria nobody knew was there.
Carbs are mandatory, never run it fasted. MOTS-c burns through glycogen while blocking new fat storage, so it needs 125 to 150 g/day of clean carbs to work. Run it fasted or keto and you starve the very engine you're building. This is the #1 MOTS-c rule.
The startup tiredness isn't failure, it's the teardown before the rebuild. Push through ~1 to 2 weeks and pair NAD+ to shorten it.
Two modes: ~1.5 to 2 mg DAILY for fat loss; a higher dose every-other-day on rest days leans it toward muscle/recovery.
Dose LOW. Anyone claiming "15 mg a day" is selling underdosed product, real MOTS-c you feel at a fraction of that.
It beats Metformin at the same job without the downside, Metformin poisons Complex I; MOTS-c hits the AMPK pathway by signaling, not by damage.
Order matters: run MOTS-c FIRST (before SS-31) so you're building a healthy engine, and prime with it 4 to 6 weeks before Retatrutide so your body can handle the fat release.

How to run itMixing, injecting, what to eat, and what to take alongside it.
Mixing and injecting
Mix: add the 2 mL of BAC water slowly down the side of the vial. Swirl gently, never shake.
Store: in the fridge after mixing.
Inject: into belly fat, ~2 inches from your belly button (thigh also works). Pinch, go straight in, push slow. Rotate the spot, fresh pin every time.
First time injecting? → Read Injection Basics (60 seconds, it barely hurts).
Your diet, this one's not optional
Carbs125 to 150 g/day of low-glycemic carbs (rice, potatoes, oats). Time them post-workout and pre-bed.
Neverfasted, keto, or low-carb, MOTS-c burns through glycogen while blocking new storage, so no carbs = a "metabolic disaster" and worse fatigue.
Whythe carbs are the fuel your rebuilt engine runs on. They're what shrinks the startup tiredness.
Take these alongside it
NAD+50 mg SubQ daily, the substrate the rebuild needs. Cuts the startup dip from ~6 weeks to ~2. (SubQ only; oral is wasted.)
Ubiquinol (CoQ10)200 mg daily with a fatty meal, the electron carrier new mitochondria can't run without.
Magnesium glycinate400 mg at night, cofactor for ATP and the AMPK switch itself.
